Detailed Description
Pipeline for Brown Treesnake (BTS) environmental DNA detection using a combined LAMP-CRISPR-Cas assay. 1. Terrestial samples are collected by swabbing multiple surfaces where BTS is suspected to be present. 2. Terrestrial samples are chemically treated to neutralize environmental components that may interfere with the subsequent analyses. 3. Loop-Mediated Isothermal Amplification (LAMP) components amplify target BTS DNA in a rapid manner. 4. In the same reaction tube as the LAMP assay, CRISPR-Cas screens the LAMP-amplified DNA to ensure that only BTS-specific targets are identified. If BTS DNA is detected, the CRISPR-Cas complex will generate a signal (e.g., fluorescence) by activating a chemical reporter. No signal is produced if BTS DNA is absent.
